SO.14.02 Two-photonmicroscopy – minimally invasive imaging in ophthalmology Oetke A.1, Steven P.1, Nuurei T.1, Hüttmann G.2, Koop N.2, Birngruber R.2, Laqua H.1 1Eye Hospital of Schleswig-Holstein, Campus Luebeck, Luebeck; 2Institute for Biomedical Optics, University of Luebeck, Luebeck Objective: Two-photon imaging is a new high-resolution optical technique that allows minimal invasive investigations of biological tissues ex-vivo and in-vivo without the use of artificial stains or fixatives. First results from ex-vivo multi-photon imaging of tissues from different species are shown to demonstrate potentials and limitations of this new method. Methods: Multi-photon imaging and fluorescent-lifetime measurements were applied to unfixed ex-vivo ocular tissues from pig, mouse and rabbit. Results: Ocular surface analysis showed corneal and conjunctival components in cellular and subcellular resolution. Whole-mount dissection enabled retinal structure analysis. Wavelength-adaptation lead to detailed differentiation of cellular and non-cellular components with tomographic reconstruction of image stacks. Fluorescent lifetime imaging allowed further cellular characterisation. Conclusions: Multi-photon imaging is a promising technique, used for experimental analysis of ocular tissue structure and function with potential for future clinical application.
